Using TrypLE as alternative for trypsin - in cell isolation from tissues (Aug/22/2008 )

Hi all!
I need help! I have a big problem with isolation of trophoblast cells from human placenta. I used trypsinization by trypsin (Sigma) and results were not good. I would like to know do you use the TrypLE as alternative for trypsin for isolation cells from tissue (not for passages in culture)? There is any difference between there in isolation technique?

TrypLE is recombinant trypsin and seems to be less aggressive than native pancreatic trypsin; try also other proteases such as pronase or collagenase or combinations

collagenase with trypLE in 2 steps of dissosiation? which of them must be 1 step?

if you have no running protocol. you have to try various protocols; we once improved aprotocol to isolate cells from a tissue only using pronase although a combination of both collagenase and pronase was recommended; the time of incubation is also very critical...

you can use two different proteases in parallel
