Tumor xenografts in athymic nude mice - re-injection? - (Jul/16/2008 )

A recent attempt to establish subcutaneous xenograft tumors in athymic nude mice by injection of cell suspension failed (i.e. 0% tumor take). Despite our extensive experience with these models, this has never happended before. We have never used this cell line before, but received it from the originator of the line who has published its use in xenograft models. After troubleshooting, we will make another attempt.

Question: What is the consensus of the group on re-injecting the same mice a second time with the same cell line? Has anyone done this before?

FYI: Original injection included 10^6 cells/mouse in 0.1 ml volume (50% Matrigel).

I would..
1. Talk with the originator (no email, unless you are too shy).

2. why same mice only? I would inject some new nude ones as well the same mice (I have never injected twice, though.)

3. Make sure that other cell characteristics follow the original description (In-vitro invasion, morphology, scratch assay etc.)

4. make sure your cells are growing well at the time of expt.

As the model in your lab works and presumably a similar model was used successfully in the lab of the cell line source - suggest you look again at what was sent. You should confirm it is what you requested as sourcing biolgical material in this manner can be risky despite the best of intentions from both sides.
Can you get the cell line from ATCC or another reputable source?
