DNA extraction - Brown color (Jul/11/2008 )

Hello Everyone,

Has anyone had problems about getting colored DNA while extracting DNA from Arabidopsis leaves? Anyone know a fast and simple method for DNA extraction from Arabidopsis???? I have never worked with plant tissues.... so any recommendations and suggestions would be helpful! Thanks.

Plant pigments can bind DNA to give a brown, green, or red color to the DNA. Adding 2-4% CTAB, PVP, and even PEG to the lysis buffer helps reduce the coloring of the isolated DNA, chelate some polyphenolics, which inhibit PCR. Some coloring is OK if your PCR goes well.

I don't know anyone has a protocol as simple as AquaGenomic or AquaRNA for DNA extraction from plant.

Yeah I do I guess. Check out this protocol we use routinely in our lab. Ok I wouldn't recommend it for cloning longer fragments or even amplify smthg larger than 3 kbp. But for standard PCR applications or cloning of smaller genes, this should be enough.
