Gel doc camera - Resolution (Jun/26/2008 )

Dear all

How can i get fine pictures from my gel doc camera?... I use canon powershot S5IS digital camera. when i use UV filter screen, i dont get clear image. Taking pictures without that screen wouldnot be a good idea for camera. Moreover, without the filter, UV lamps are prominent and gel appears in the background.
I use following specifications in the camera
Mode: Tv
Shutter speed: 15sec
ISO: 1600
Color mode: B/W
ummh what else.??..

Pictures are not good like those seen in google. I find pictures dark and there are dots in the pixels when visualized in computer even though the picture size is more than 3 MB

Can anyone suggest me any change in my operation??

What about focus? You use auto or manual?

hi,

your shutter speed is higher than what we use (at most 3 second)
check the fucosing first then use lower ISO for darker picture ( it removes backgrounds)

The mode is Tv mode as the manual for camera suggests...Its a form of manual mode.
Most of my friends in other labs use auto mode to take pictures...

With my settings it works well but its a bit time consuming... it takes 15 seconds to take a single picture...

What settings do you normally use?

"Tv" mode is manual shutter time and automatic iris (aperture), that's have nothing to do with focus. Digital cameras have problem with focusing on blurred objects (eg. electrophoresis bands) and surfaces with no distinct details (eg. transilluminator). Tell me, do you see focus "box" when you take a photo? Green or yellow?

S5IS is not easy to work with for lab work on gel image.
It took us a while to set it up and we still don't like it.

I will try use smaller size of picture that will reduce the size of file. I don't think you need that big of picture, on computer even 600X800 is a little too big.

As I can see, you might get ISO too high, higher ISO you can shorter the time of shutter speed, but the quality of picture will reduce as described in your message (dots in pixels).
The one we are using ISO 200 seems to be enough for my purpose.

One thing make sure no light leaking in the hood so the contrast between bands and background will be greater and it can help to reduce the sutter time. I using 1.5-3.0 sec currently, but we need turn off the light in the room.
For same reason, don't put too many EtBr in gel it will reduce contrast.

I've found some filters give much worse pictures than other filters that seem pretty similar. I can't say i know too much about this but it's something you could try - a different filter.
