Arabidopsis agro infiltration - (Jun/18/2008 )

Hi.

I am trying to express transiently some proteins in arabidopsis leaves....But it seems that it isn't as easy as it is for nicotiana...I tried different protocols for this, but still can't get it to work....
I think that incubation time of agrobacterium is important (I incubated them in LB ,plus antibiotics and acetoyringone (50ug/ml)).I icubate them for 2 days...After that incubate in infiltration media (MgCl2 10mM) or just destilleted water plus antibiotics and acetosyringone for 5 h. wash twice and infiltrate 3 weeks old plants.
Does anyone know if there is something special about arabidopsis and Agrobacterium that is different for nicotiana? I am sure there is but a little bit lost about what can it be.......

Any advice is really appreciatted! blush.gif
