MCF-7 & MTT assay & E-screen - (Jun/18/2008 )

Dear friends

i performed the MCF-7 PRE assay in 96 wells.. however when i counted the cell viability using MTT assay, i found that the formazan was not attached on the bottom of wells.. so, i tried to sentrifuge it (3000rpm, 5 minute).. but its still not attaching.. is there anything wrong with my experiment?

beside that, my MCF-7 cells are really strange nowadays.. they have a very long dendritic cells.. i bought that cells from ATCC and i used DMEM as the maintaining media (following most paper methods).. is there anything wrong also with my cells? how to make sure that my cells are really MCF-7? is pS2 protein a specific protein in MCF-7 cells? please help me...

Dentritic in MCF7?....impossible....sometimes cells change their shapes due to condition of the bottom of the flask and using different flasks from different companies....that might be the issue.....this happens to me too....sometimes my HeLa cells have a different morphology.

there are some filter columns in the market that are for dividing different cell types....I'm not sure though, you need to check, we have a kit and column called MACS or something....it's from Axygen.
