EROD Assay procedures and solutions experts please help - (Jun/10/2008 )

Has anyone here done EROD assay? I am writing a protocol for EROD assay, and I have a few things that need to be cleared up.

We have 7-ER powder in our lab, and we are trying to make a stock solution from it. But I couldn't figure out what concentration might be needed, and various articles indicated that people were using different concentrations for different experiments. so how to be sure which concentration to use?

I read some articles in which people use dicumarol (an NADPH inhibitor), what is the true purpose of it?

Is NADPH necessary in EROD assay? From I have read, it is needed to initiate the reaction, but some articles have no mention of it even though they are doing EROD.

If you have any additional input as to what is important in doing EROD, please let me know.
