CHIP : Use frozen tissue - (Jun/05/2008 )

Hello. I am trying to set up CHIP experiments using plant tissues.
I have to use frozen tissue and I dont know if I have to defroze it first and then put it to formaldehyde or if I should put it to formaldeyde right away. Do I have to extend the time that the tissue stays in formaldeyde solution if the tissue is frozen?

I am not using plant material, but I am using frozen cells for my ChIP. With our cells we thaw, then culture for 24 hours before doing the ChIP. Having said that though, we have thawed the cells and immediately gone onto cross-link...the results were the same as the cultured cells, although we recovered much less DNA.
