How to detect the presence of Poly(I:C)? - (May/14/2008 )

I am using Poly(I:C) as virus mimicry in the IFN experiments. I wonder if there is any method by which I can show Poly(I:C) does enter the animal. For genuine virus, I think I can use PCR to amplify a species specific sequence. But what can I do for Poly(I:C)?
Any suggestion is welcome. Thanks.

I don't think anybody does Poly(I:C) assay. But if you want to cover everything, I guess you can probably do interferon levels. If interferon levels are up, it would be an indirect measurement of your Poly(I:C) function. For IFN assays, see below:

Actually, I am trying to detect the level of IFN and its target genes in my experiments. I want to show that the level change is due to the introduce of Poly(I:C). So if I use IFN as indicator of Poly(I:C) presence, it is kind of circular logic.

Well, if you use Poly(I:C), you are bound to induce interferon. Irrespective of your genes and targets. But it may be diffferent in control Vs. Test mice.

As long as you have vehicle treated mice and p(ic) treated wt mice as controls, I don't see any problem. Anyways, that's life! Have fun. tongue.gif

You may try this method to label it with 125I.
