the difference between YPD and SD - (Apr/23/2008 )

hi- maybe someone here can help me solve a Q' i got on a microbiology class:

I have grown EMS mutated yeast on replication plates of YPD and SD.
some of the colonies appeared only in the YPD...
Assuming that i tried adding all of the nutrients known to man (!!!) to the SD -and still they didn't grow- what can be the reason for the difference?

1. i need to try a combination of nutrients because more than one might be necessary.

SD (synthetic deficiency) medium consists of a nitrogen source, Glucose, amino acids and water. To weigh all these amino acids is a really time consuming and annoying work but in some cases not enough to give your cells a cosy and nutritious surrounding. In the Yeast extract and Pepton which you add to the YPD medium there are much more nutritions supplied than in SD.
