Calcium Flux by FACS using Fluo-4 - Can the cells be fixed? (Apr/14/2008 )

I am going to use Fluo-4 dye to measure cell calcium flux. I was wondering if the cells can be fixed before FACS measurement. Will the fixing affect cell calcium release or Fluo-4 dye?

yes, do not fix your samples for Ca flux analysis. you need live healthy cells in order to get a true read-out of the signaling that is occuring in your cells.
