Having trouble counting PBMCs - (Apr/11/2008 )

Does anyone have any tricks or advice for counting PBMCs with trypan blue staining? Since it is a pool of different kinds of cells, they are all different sizes and shapes. Some are so small it is hard to tell if it is trypan blue stained or not. Would it be better to just count one cell type and use that as a base count for the pool as a whole? Would a different counting method be better?

in our lab, we only count the round cells that look nice, not with strange shape, not too big.
