looking for HCV positive cell lines? - (Apr/01/2008 )

To work with our project, we need a HCV postivie cell line, or at least it can express the core region. would anyone know about this kind of information. I searched online, but did not find it.

It is not clear from your post as to what actually ur project is so i wont be able to provide a solid opinion. as far as my own knowledge is concerned, you will have to develop these cell lines by yourself.
can u plz tell me the details of your project and why u need the cell line and specifically on which genotype/subtype ur working caz therz a lot of heterogeneity (as u might be knowing) in HCV and proteins of one genotype/subtype do not work against the ones of other types specially if ur going to carry out some immunoprecipitation etc kind of experiments using Abs in ur study.

I agree your question is not very clear, but this might help. This group has developed a cell line that produces HCV-like particles, none of the genetic material, but the same proteins in the viral envelope.

It is not clear from your post as to what actually ur project is so i wont be able to provide a solid opinion. as far as my own knowledge is concerned, you will have to develop these cell lines by yourself.

Actually what I need is a cell line, better to be cancer cell line, which is HCV infected, whatever kind of genotype, as long as the core region is expressed in the cells. becaues core region is quite conservative among different genotypes of HCV. I need to evaluate the immune response of CTL toward HCV core region expressed target cell lines by 51Cr-release assay .
so if HCV infected cell line is available, that will be the best. if not, then even HCV core region overexpressed cell lines will also be useful for my project.

you primed your CTL with wat peptides? or you have or going to isolate it from some infected patients blood?
i still guess it would be difficult to find such cell lines but i will check and i think i came across some such stuff on web a few days back. but im not sure if the cell lines i read abt were expressing core or wud they be useful for u. i will check it again

as for the conservation of core, its a little cliche i guess. my own analysis with the protein sequence alignment of subtype specific consensus sequences reveals that among the most common genotypes (1a, 1b, 2a, 2b, 3a) the conservation in Core region is roughly 84%. this might be a high conservation when looked at it in the context of HCV but still u need to consider it regarding wat peptides u used to prime ur CTLs or the infecting genotype of patient from whom ur going to isolate ur CTL.
