criteria of the parameter that is used for normalization of RLU data - (Mar/15/2008 )

Dear all
I have one inquiry about the criteria of the parameter that is used for normalization of RLU data of luciferase assay, should it be NOT affected by drug treatment? I mean like the case of internal standard in real time PCR for example? because in my case, drug treatment reduces protein concentration because of the S-phase arrest it induces, so could protein concentration be used as a partameter for normalization of RLU data?

I've read the other post you have in this forum. Maybe you could try to use other normalization method, for example, cotransfection of a Beta-galactosidase expression vector (for example pSV-BGal) in combination with your luciferase reporter plasmid and then you can normalize your luciferase activity against B-gal activity.

Anyway, I've read a lot of papers with protein normalization of RLU after treatment with different drugs that regulated cell cycle or were cytotoxic, but is real that you must choose an internal control independent of the treatment.
