normalization of luciferase RLU data - (Mar/13/2008 )

Dear All
I have done luciferase assay for a drug that induces S-phase arrest in human cancer cells. values of the RLU (raw light units) in drug-treated samples were similar to those obtained with control vehicle treatment. when I normalized the RLU values to the total protein content, I got a statistically significant difference between both sets of samples, since in the drug-treated samples there were less protein content. I am confused about interpreting the results whether there is a transcriptional activation of my target gene or not.

Yes of course you need to normolize to total protein conc. if there is a difference, yes there is effect
