Non-growth of E. coli TOP10F' in LA+tetracicliyn media - (Feb/28/2008 )

I transformed some E. coli TOP10F' cells with pUC19.
The transformed cells were plated in LA+tetracyclin+ampicilyn and i had some growth but i coudn't see any satalite colonies.
My control cells that were plated in both LA+tetracyclin and LA+tetraciclyn+ampicilyn didn't grow. I didn't have a single colony!
In both cases the cells were incubated at 30ÂșC for 30-40 min in LB.

Can somebody help me find an explanation for the lack of growth??

Your Top10 F' cells should grow on the tet only plates. If they do not, they either never had, or have lost, their F plasmid. Are you sure they are F' rather than plain Top10?

They were provided by a professor and he garanteed the presence of the F plasmid. Besides the transformed cells came from the same culture and they were able to grow with tet present.
