Different ways of differentiating actin and myosin? - (Feb/19/2008 )

Hey,

what are possible ways of analyzing the composition or structure of actin and myosin to see the difference between them.

I already know one by using Bradford protein assay and SDS-page, but I was wondering what other methods I could use?

electron microscopy of the filaments (negative stained).

Would that be similar to fluorescence microscopy. Basically staining actin or myosin and looking at under electron microscope or fluorescence microscopy.

Thanks for the reply.

not fluorescence microscopy.

we would prepare actin and/or myosin filaments and send them to the em lab. there they would apply the filaments to grids, stain with uranyl acetate (negative stain) and then look at them with the em.
