How to isolate gastric parietal cells? - Do you have protocol for isolation gastric parietal cell? (Feb/13/2008 )

Dear All,
For my experiment I need gastric parietal cells. I did not fine the cell line of this cell in ATCC or ECACC Catalogue, therefore I plan to isolate this cell from Rat Stomach . Does anyone know how to isolate gastric parietal cells? Does anyone have the protocol for isolation gastric parietal cell?

there are 2 classic protocols which are modified by various scientists; for rabbit you should do high pressure perfusion (as for hepatocyte isolation) for rabbits with elutration;

for rats and mice do the classic Lewin protocol; modifications use a cell strainer;

both protocols should be finished with a Nycodenz step gradient

you need Lewin A, B and C; follow the instructions to be read in:
93. Lewin M., Cheret A. M., Soumarmon A., and Girodet J. (1975): Method for isolation and separation of cells from rat fundic mucosa. Biol Gastroenterol (Paris) 7, 139-144.

it is hard work; I advice to spend a spell in a lab where PC are routinely isolated...
