Fluorescencece average - (Feb/13/2008 )

Hello, I am a new user for FACS analysis

So,

I am doing macrophage's infection with fluorescence expressing mycobacteria. I use different treatment on my macrophage before infection. After infection I use FACS analysis to quantify the percentage of phagocytosis but I would like to know if it's possible to know the fluorescence's average per cell. It could be interesting to see if my treatment can increase the quantity of phagocytised mycobacteria compared to a control without any treatment.

FACS is good; inspection with fluorescence microscope will specify the events;

is Mean Fluorescence Index a regular term? what does it exactly say?

Oh, Thank U bearer for pointing that out. It was my mistake, I should admit. I meant to write Mean Flourescence Intensity (MFI).

But, I have come across the term flourescence index also in some papers. I used to be really confused what actually is 'MFI'.. . Mean or Median and Intensity or Index. I still have that confusion. I had asked about whether using 'Mean' or 'Median' in one of my posts before.

About the term 'index' please give me some time to find the article. I am engaged with some work. As soon as I get time, I will find it. Googling didn't help. I have to search the pile of my collected article. No search engine works there - just a pile of paper in my drawer. sad.gif

Sorry, The Bearer, I could not find the article I had consulted but I found simialr use in another ( http://www.jci.org/articles/view/3714/pdf in methods )

It is the ratio of difference of sample mean fluorescence intensity of sample and that of negative control to mean fluorescnce intensity of negative control.

I am still confused of the terms - Mean fluorescence Intensity, Mean Flurorescence Index, and Mean Fluorescence Intensity Ratio (MFIR). Can any one please throw light on this.
