How to create a mutant DNA having point mutations sequence? - (Jan/21/2008 )

Hello everyone,

I need to make a mutant DNA from a cDNA. I want to have change of three bases so that a different amino acid is expressed, in other words I want to create amino acid point mutation. Can anyone guide me through this? Suggestions, links, protocols etc all welcomed.

Have you looked into trying side-directed mutagenesis? Search the forum and also stratagene has lots of info about it.
