Bisulphite sequencing - Fwd sequence (Dec/19/2007 )

Hi,

Does anyone know why the fwd sequence tends to drop in signal strength/bad sequence quality? I use M13 tails, but doubt that being the problem.
I notice that other people have the same problem, but don't understand the reason...???

I think it's to do with the base composition of the synthesised strand.

It's very AT rich with some G thrown in for good measure. The poor quality is due to the Taq struggling with poly AT runs I suspect.
