how to do intratumor injection? - intratumor injection (Dec/11/2007 )

hi, i want to establish tumor xenograft model, and then inject the plasmid DNA into tumor, but i never did it before, so i want to know how to do intratumor injection, and when to begin (also how much size of tumor ), someone said tumor can be injected directly, however, other said the injected site should be far away from tumor, the needle should slip subcutaneously, and then into tumor. i am puzzled which is right.

another question: compared injection by tail vein with intratumor injection, which is more efficient? and if inject by tail vein , how much plasmid DNA can reach tumor xenograft?

please help me. i will appreciate your help. 3ks.

tail vein injection of pure DNA is ineffective. i.t. injection of pure DNA? maybe, but weak. i.t. with electroporation worked. Some used i.v. DNA complexed with PEI, DNA with PEI and albumin, liposome or polymer-transferrin conjugates showed positive results.

hi, genehunter-1, thank you for your answer. you said some used i.v. DNA with liposome or polymer-transferrin conjugates showed positive results. have you done it before? if compare i.v. DNA with liposome or polymer-transferrin with i.t. DNA with liposome or polymer-transferrin, which is more effective? have you read some related paper? can you give me some suggestion? thank you again!!!
