Keeping IFN in cellular media - (Nov/30/2007 )

Hi,

I have a question for you.
I prepared some media (RPMI 1680) with IFN and keep it at 4°C. How long can I keep my mix without any degration of interferon??

You would have the same amount of time as if you diluted it and then put it in the refrigerator. You only mix it when you use it, just like you only adjust the pH of a solution when you use it. You're screwed. ohmy.gif

Dear Biofred,

We use IFN-gamma in combination with LPS to stimulate J774.A1 (macrophages) to produce inducible Nitric Oxide Synthase (iNOS). We normally aliquote the IFN into smaller amounts to reduce the number of freeze-thaws it undergoes i.e

100,000 units in 1ml .....aliquoted into 50/100ul amounts.

You should always use fresh IFN for your experiments....NEVER add it to RPMI and then store at +4oC.....it will drastically lose it's activity. Our IFN is stored at -80oC and is active for upto 2 years.
