Lipofectamine LTX - (Nov/20/2007 )

Hello,

Has anyone used Lipofectamine LTX from Invitrogen? The manufacturer's claim that its better than FuGENE (in terms of cytotoxicity induced and percentage of transfection). I use FuGENE and have got some very good transfections with it. I wish to know if Lipo LTX is better. I need to get a transfection reagent and Roche isn't currently in the rate contract of our instuitute whcih is why I am forced to look for other sources. Any suggestions??? Thanks in advance

Ask for a free sample and try it. (btw: Roche claims that Fugene alters expression of a lot less genes than Lipofectamine, so it's most likely commercial talk of both companies).

Maybe specifiy what cells/cell lines your working with, optimal reagents may differ between different cells.

its all advertisement. Choose any reagent that you have optimized for your cells.

we do neither use LTX or HD if we have to transfect genes of cytotoxic proteins; Ca-PO4 with DMSO and chloroquine-step is still a very good alternative

The best which I had ever used. I am not attached to Invitrogen in any way but I carry 6 different cell lines and lipofectamine LTX consistently gave the highest transfection rate for all my experiments. I had tried:

1. Lipofectamine 2000k
2. Fugene
3. Verofect
4. Dreamfect Gold
5. Amaxa (electroporation)

Different cell lines give different transfection rates at different seeding conditions and exposure time (serum free/ with serum), DNA/lipid concentration

With LTX, you can also try PLUS, nupherin, combiMag, etc. Variations are endless (but your time and budget is smile.gif ).
