good positive controls for Chip - (Oct/30/2007 )

hi,
I have to do Chip to check whether a transcription factor can bind with the promoter of a target gene. I guess I should start with a good positive control to make the Chip system working. Could anyone please give me some advice for the best positive control for a newbie to start with? A system in a murine cell line will be best for me.

Thanks

I think for validation of system histone proteins are good to start with.

polII is a good candidate for such a thing it is used as a positive IP control and you just need to pick an actively transcribing gene such as a housekeeper to test the system.

Thank you very much. I might have a chance to get the protocol and reagents for Chip between pol II and b-actin as well as another gene from another faculty. I will keep posted after I work everything out .
