Nystatin makes vero cells round, too high concentration? - (Oct/29/2007 )

I have no experience in using nystatin. The concentration of nystatin in my experiment is 50ug/ml. 80%-90% of Vero cells became round and detached from the flask.
1.Is the concentration too high?
2.I think, for the cells becoming round and detached, nystatin worked effectively, and for those with the nomal shape and still attaching to the bottom of the flask, nystatin did not work effectively or worked less effectively. Am I right?

What are you trying to do? Measuring LD50 in Vero cells for this drug or use it to kill fugus?

Thanks! I use it to disrupt caveolae of the cells.
