Activation of JNK signaling pathway using frozen liver tissue? - (Oct/11/2007 )

To anyone who has had experience working with JNK,

Is it possible to probe for P-JNK using frozen liver tissue? I do not know much about this signaling pathway, but from what I have read it is activated by different forms of stress. Most of the protocols in the literature have treated the animals in order to induce the signaling activity. Then they proceed to sacrifice the animals and collect the liver for analysis. Any feedback is very appreciated!!

what is frozen? cryosections of liver tissue? then it will be possible, but if you are not experienced with IHC, do it in cooperation with an expert

I harvested the whole livers and stored them in -80. I took a portion of the liver sample, homogenized it and then determined protein concentration

using whole frozen tissue is highly critical for desintegration of lysosomes and caused proteolysis when thawing; you have carefully to check protein degradation
