direct vs. indirect labeling in microarray exp. - (Sep/28/2007 )

Hi all,
I am new in microarray filed, and want to know what the differences between direct and indirect labeling are. On which basis can i choose one of them for my experiment?
Thanks glare.gif

Hi,
first of all in Direct labelling Flourescence labelled dNTP incorporated in cDNA during polymerase reaction and in the case of Indirect labelling first of all cDNA or RNA synthesised and then a dye cuopled to incorporated nucleotide.
The main advantagese of indirect labelling is high efficency of attching of flourescence tag after labelling and in the case of direct labelling minimmum effecency of incorporation of modified dNTP during cDNA synthesis.
According to my thinking indirect labelling best for microarray experiment.
