TCA radiacive protein precipitation without lysis - (Sep/17/2007 )

Hi,

I know from literature that it is posible to precipitate proteins with TCA from total cells- without lysing them before. But I cannot find any protocol for this procedure. Probally anyone can help me.

Up to know I have always lysed the cells in buffer with Triton X-100 or NP-40 before TCA precipitation. But now I need to precipirate radioactive proteins imiedetelly

it is a harsh method of lysis as proteins become precipitated, and you have to work with highly acidified lysates; use the same conc of TCA as for protein precipitation; work with cooling
