seed sterlization question - how long do your seeds last? (Aug/26/2007 )

Hello,

I commonly do seed sterilization on A.thaliana seed lines. I was told that if I don't use all the sterilized seeds, that I can store them in the fridge and they will last up to 2 weeks. After that, they may not work right.

I tested this theory out with seeds that were sterilized a month ago and were stored in the fridge. I plated them on regular MS media, and they grew just fine. So now I am wondering, how long do seeds that have been sterilized typically last? Thanks.

i don't know, but i think they become dormant in fridge conditions, and by providing them with suitable conditions, they can grown again easily....but b sure they not contaminated !

Well first off i better say i cant answer your question, but you may be able to help me. Can you tell me where i can find info. on how to make the culture media for A.thaliana?? What is the 'regular MS media you mention??

Iam dealing with seeds of Cicer arietinum. I am new in this field I have tried to sterlize my seeds by Na hypo chlorite; SDW thrice and air dry in sterile conditions but still fungal contamination occurs. Cud you help me in telling me how to sterile the seeds.

Hiya I am new to this area too, i have read in a few papers the method you use, -


Surface sterlisation: with a 1%(w/v) hydrochloride solution for 3 minutes and then kept in fridge at 4 degrees or a lot of papers just say the USEPA method they imerse the seeds in 10% sodium hydrochlorite fro 10 minutes.
but the method i am going to use for A.thaliana is to surface sterlise with 70% ethanol for 5 minutes, followed by soaking in 0.5%(w/v) na dodecyl sulpahte ( SDS) solution for 15 minutes and then rise with sterile water, i will use them immediately after that, and not dry them, just place them in my sterile growth gel.

hey NK 111 if you find teh answer to that question , i would be interested to know smile.gif I would liekto keep a selection of my seeds and wonder how long they will last.

I know that I store seeds in the refrgeratory with 4 degree for more than 1 year, and they can work well.

I often store seeds after gathering and do not sterilize them. Actually, seeds are only sterilized just before germinating on the plate with 1/2 MS

hi guys,

first of all, i'm not working on arabidopsis, however, if i'm not mistaken, arabidopsis seeds need some time at 4 degrees to break dormancy and have a better germination. you shouldn't have problem at storing your seeds in a fridge, for how long you can store them also depends on how long ago you harvested them.

I've germinated seeds that were sleeping for about 10 years in the fridge (Nicothiana spp).
Seeds are really though things, so, as they were conserved correctly I gave them a try.
I sterilized them before plating, because I didn't know how they were treated and obtained a fair number of plant in about 15 days.

I don't know how different it can be with Arabidopsis seeds.

They can last for years, but gradually lose their fertility with time. If you keep them in a cool, dark place in a box containing dessicant they last best. Mutants may vary.

MS is 8 g agar and 4.24 g Murashige and Skoog salts in distilled water, adjusted to pH 5.7 and autoclaved. You can add other nutrients as well if you want - a lot of people add 3 % (v/v) sucrose.
