Is that possible to establish a stable cell line which is transfected by an empt - (Aug/21/2007 )

Such as a cell line transfected by pcDNA3.1(+).

I'm an idiot on establishing stable cell lines sad.gif
Will the integrated empty vector express neomycin so that the transfected cells survive the G418 toxicity?

Thanks,

yes,

if you establish a cell line expressing a specific protein you would also need a 'negativ' control cell line-undergoing the same procedure (transfection, selection, integration...) like your protein expressing cell line. How can you otherwise analysis effects associated to the protein expressed in your stable cell line.

Usually these vectors contain two promotors - SV40 for the resistancy and an CMV promotor for the expression of the protein.

Thanks, Bomber

I was just a little bit confused when my cells transfected with empty vector also survived the G418 selection.

Thanks
