cell density for infection with baculovirus - what is the optimal cell dentisty at the point of infection (Aug/13/2007 )

Hi,

I was wondering what is the optimal cell density at which one should infect insect cells with recombinant baculovirus for protein expression. I grow sf9 cells in suspension in SF900 II SFM supplemented with 5% FBS.

I have expressed 3 isoforms of Nitric oxide Synthase in both sf9/sf21 cells. In my experience 90%confluence of what ever flask/dish you have is OK. The most important thing is the Multiplicity of Infection/plaque forming units i.e. the MOI/PFU. For all my isoforms I found that the best MOI/PFU is 5 i.e. 5 virus particles/cell.
