do we need to wait before doing mtt? - (Jul/11/2007 )

hi,
i need to do mtt assay for ht29 which is treated. some of my friends said that after putting 10000 cells into each well, we need to wait for at least 24 hours before we add in the compounds which i am desire to test. is this necessary? they call this as seeding...cant we do serial dilution and then put in the cells straight?

Well, it is right. 24 hour incubation period is necessary for cell monolayer (middle density) formation. For each cell line it is very important to know start concentration of cell suspension to reach after 24 hour suitable density of cell monolayer.

cells are stressed after splitting, and need time to recover and to get adhere; waiting for 24 h is necessary; in this time, cells poorly proliferate
