Identification of bacteria from marine fish. - (Jun/24/2007 )

I'm doing a project on isolation and identification of bacteria from marine fish.

Right now, I've been at the stage of storing the bacterial culture in 80% glycerol solution in -80 degree Celcius.

My questions are:

1. Do I have to perform Gram staining to all my cultures? (I have over 100 cultures) Or should I just do the API test instead?
2. If I were to skip those tests, Can I proceed to 16s RNA identification protocol?

I really have no idea. I've been doing a lot of readings and it is getting confusing.
Help me, tq so much.

As far as identification is concerned, though the phenotypic way of identification is laborius and time consuming, it is more reliable... It is better u do Gram staining and other preliminary identification tests like the motility, catalase and oxidase before u proceed to the complete set of biochemical anlysis... Though 16s RNA identification is precise, it depends a lot on the purity of the isolates u have... Its is costly and moreover 16s RNA identification is dependent on universal primer, hence less reliable. So first narrow down with the phenotypic identification...

Nb: U have mentioned 80% glycerol solution... Is such a high conc. of glycerol necessary?? sometimes the glycerol can be lethal to the organisms... plz refer...
