Thp-1 cell transduction - (Jun/22/2007 )

Hi
I'm trying to transduce THP-1 cells using lentivirus. Does anyone have experience of transducing THP-1 cells I'd gladly accept any tips or advice on the topic smile.gif

Hi Susie,

If you're trying to transduce THP-1 cells using lentivirus, I advise you to use ViroMag R/L, which is supplied by OZBiosciences. ViroMag R/L is a magnetic beads containing formulation specifically designed for retrovirus and lentivirus, which allows to increase your transduction efficiency. Actually, you just have to mix ViroMag R/L and virus to be transduce and then vectors and viruses associated with magnetic particles are driven into target cells using a magnetic fields. It works very well.

Actually, several papers show a significant increase of transduction efficiency by using magnetic force to drive viruses & vectors in association with magnetic particles into cells. Personnaly, I use Magnetofection to tranduce my cells because I obtain better results with it.

choose promoters in the lentiviral vectors which have been proven to work in these cells.
