problems about THP-1 cell culture - (Jun/21/2007 )

this is my first time working on a suspension cell line. yesterday i centrifuged my THP-1 cells, removed the medium and add serum free medium for serum starvation. did i do anything wrong? or is there anything should be especially teken cere of?? bc after that, i found the cells started to adhere><" seemed like i started to differentiate into macrophage like cells.....hope there's someone to help me. thx!!!

These cells normally grow in 10% FCS.......... these cells will not be happy in serum free conditions. If you reduce the serum slowly over a long period of time then they will be more able to survive serum starvation. The other thing to find out is:
What were the cells frozen down in? They always need to be frozen in Glycerol as DMSO will Differentiate the cells.
It sounds to me that the cells HAVE differentiated into macrophages because THP-1 are suspension cells and Macrophages are adherent
