siRNA transfection - (Jun/21/2007 )

Hi,

I am working on primary mouse macrophages. I need to transfect siRNAs into them. Could anyone help me with regard to the protocol and suggest the ideal transfection agent I need to use?

Hi !

I have never tried to transfect primary macrophages, but I have ever transfected SiRNA into raw264, which is a macrophages cell line with PolyMag from OZBiosciences and it works. PolyMag is a transfection agent based on the magnetofection technology developed by this supplier. You can take a look on their website for more information.

Thanks but I am working on mouse peritoneal macrophages that are more sensitive to transfection agents than that which can be used on RAW cell line....so I am still trying to figure out what can be used as an optimal transfection agent...any suggestions on that?

Primary macrophages are very tough to transfect. You may try electroporation or lentiviral vector.
