G418 and hygromycin selection question - (Jun/20/2007 )

Hi ,
I have already established a G418 resistant cell line (HEK293). I want to do a second stable transfection using hygromycin selection. When I start the hygromycin selection do I also include the G418 in the medium or can I only have one antibiotic in the medium at a time?

I would still keep G418 in the medium, albeit at a lower concentration (the maintaining concentration) as compared to what you used before for selection. That would help to ensure that your first stable gene is maintained.
