what is the best dna methylation/modification kits? - newbie kit comparison (Jun/12/2007 )

Hi all.... im fairly new to the dna methylation analysis/dna modification community using kits, and would like to give a tryat kits instead of homebrews

if anyone has used kits before, can someone tell me how well the results are (and ease ofuse) compared to traditional homebrew recipes?

also.... ive been lookin at kits online and came across three im interested in:


ezdna methlyation kit (zymoresarch)
methylamp dna modification kit (epigentek)
methyleasy dna bisulphite mod kti (humangentic signatues)

im really intrigued by the zymo and epigentek kits in particular, the steps look really simple -- but is it too simple and will be less effective than home recipes? ive been perplexed by the epigentek kit as it looks like it has alot of great features really cheap but i wasnt able to find many references for them on google (i guessthey are new or something?)

anyways could someone who has experience who have tried the above kits tell me the advantages and disadvantages of each, or what you recommend?

would appreciate any help!

I use Qiagen's Epitect kit which is very comfortable. No need to adjust any pH, very easy steps.

I use the modified DNA for MSP and direct sequencing, both work excellent, sequencing confirms good conversion.

I´m also using the Qiagen-Kit and i get very good results. It´s works great for me...

thanks...how long does the qiagen kit take (or more specifically the reaction time?)

The reaction time is about 5 hours. Columns for clean-up are included in the kit. Thus the conversion is not very time consuming!

I have tried ezdna and methyleasy, both work well, however the yield from each is drastically different.

With ezdna you lose the majority of yo9ur DNA after bisfulte like with the homebrew method. Methyleasy does not degrade your DNA.

Ezdna is very easy to use, methyleasy is easier than homebrew method but is not column based.

hmmm... I'd love to give methyleasy a try but I just realiezd the price is a little out of our range for 25 reactions, especially for me to try kits for the first time.

I think I will order either the Zymo or Qiagen kit, will update here.

We had Zymo and Qiagen, the latter working better in my hands...

Thanks, good thing I decided to order a Qiagen kit... will try it and let you all know how it goes

we use zymo exclusively. every person in the lab can get it to work.

i'd prefer to try some of the other ones out, but once you have something that works, it's impossible to get this people to change.

we use it for general Bis PCR, clone, sequencing....
