Can cell culture media be frozen at -20°C? - (Jun/05/2007 )

Did anybody of you ever freezed the cell culture media at -20°C before using? Is it a problem for cells (specially for fibroblasts) to grow with this?
(Of cause I will warm it up before using cool.gif )

I think it's ok. Since serum can be frozen, medium containing only small molecules should also can be.

I think it's ok. Since serum can be frozen, medium containing only small molecules should also can be.


Ask yourselves " How is the media delivered to you, Frozen or liquid" ?

What do the MANFACTURERS state on the bottles are the correct storage temperature ?


If you only use a small amount of media then only make up 100ml of complete media. It is not recommended to freeze complete media.

the fcs we aliquot and freeze, same with the pen/strep and trypsin/edta then you just add what you need to the medium which you then keep in the fridge

Normally, media is always kept at 4 degrees and not stored frozen. Also because some of the amino acids present in DMEM/RPMI degrade when they are kept frozen for long such as glutamatine. Make aliquots of 100 ml and may be store and use them within 2 weeks.

If it is kept sterile, culture media can be stored for long periods at 4 celcius. You just have to add glutamine if you store it for more than a month.

I totally agree Madrius, good post. However how many times have you been to the fridge and found media that was 6 months-1 year old?
