Modification of Hematopoietic stem cell - (May/08/2007 )

Hello!

I have a theoretical question.

Hematopoietic stem cells (HSC) are stem cells and the early precursor cells which give rise to all the blood cell types.

We transfect these cells with lets say one additional gene.
I want to change all the blood cells with the aditional gene. It should take a lot of time before this happens(60-90 days).

I also know that gene therapy is still not succesfull enaugh. Lets assume that efficency will get better in future.

The main question therefore is: Can we change the majority of blood cells by changing the hematopoietic stem cells.

please, note that in this time were any findings from Christoph Klein, Medicine Faculty,Hannover. Wiskott-Aldrich-Syndrom, about this disease is the research too. WASP-Gen was genetically modified.

what about CD34+ cells, how do you culture this cells? with methyllcelulose or w/o

Actually, I'm trying to expand CD34+ cells in vitro using a serum free medium completed with L-glutamin, SCF, TPO, Flt-3 and serotonin.

Yes, I have medium with cyokines SCF, TPO, Flt-3 and GM-CSF, IL-3 (what about that?)
I prepare my cells with MACS Technology.
But I take for my medium yet serum. I know, it shall be not so good.
Please any advice.

My MACS Vario magnet has absorbed some moisture on one of the inner sides behind one aluminum cover and expanded outwards about 2mm. See attachment. Do you know if this magnet is still good.
