G418 effect on protein concentration - (Apr/06/2007 )
Hi,
I am growing mES cells with different mutant constructs, some of which are in G418 media, and some without G418. When I did westerns on some of these cells we noticed that if the cells normally grown in G418 were grown for 2-3 passages in media without G418, their concentrations of certain proteins were different. There was more protein in cells grown without G418.
Has anyone else experienced these differences, and if so, can anyone direct me to papers that explain the mechanism behind it?

beside the specific resistance factor against G418, other detoxifying pathways such as multidrug resistance proteins might be activated
