Culture medium for MDA-MB-231 / MCF-7 - which media is good enough? (Mar/20/2007 )

Hi,

I'm trying to get a culture of MDA-MB-231 / MCF-7 cells going but can't really decide which culture is good enough.
ATCC recommends L-15 medium and some special MEM cocktail respectively, but I've seen several papers which use RPMI 1640 or DMEM for both.
If the media doesn't affect the growth that significantly, I would prefer RPMI 1640 / DMEM since I have a lot of stock for these media.

From your experience, are there any problems with RPMI 1640 or DMEM with both cell lines? If not, which media is better for each (i.e. RPMI 1640 or DMEM), and are there any adjustments I need to add to the media? (ATCC recommends adding some insulin to media for MCF-7 cells).

ive always used RPMI 1640 w/ 10% FBS, 2% P/S, 1.25% L-glut, and 1% sodium pyruvate with absolutely no problems

L-15 is designed to be used not in a CO2 controled incubator

I use DMEM+10%FBS +NEAA for MDA-231 cells and it works great.

We use high glucose DMEM (4.5g\l) 10% FBS, EAA, NEAA, 2mM Glut, and piruvate + antibiotics . Work good!

About MCF-7

This tumor cell line was derived from gormone-dependent mammary tumor. So instead of MDA MB 231 you should add insulin to cultivate thi line! It is a rule for this tumor cell line. A question is what quantity of insulin you need! It is depends on your exp and the rate of proliferation you need. Typically conc range between 3-10uM.

Circlepoint, thanks for the info about the insulin. So will D-MEM or RPMI work well for MCF-7 as well?
Also, should I use insulin from human, or is it ok to use insulin from bovine pancreas (that's what I have now)?

Hi,

so everyone agrees about the possibility to grow MDA-MB-231 (and other MDA lines...?) also in DMEM or RPMI...but I have not understood well if it is necessary to add insulin and why it is/is not?

I am just starting to culture them and in tha lab they say to simply use the same medium as for MCF-7 cells
