HL-60 CULTURE - Hematopoietic stem cells (Mar/16/2007 )

I am looking for a protocol to for HL-60 cell ine culture.

I culture them in RPMI 1640 with ultraglutamine + decomplement FBS 10% + Atb, only

i culture HL-60 in RPMI + 10% FBS, P/S and they seem happy. I usually seed a T-25 around 1x10^5 cells/ml and passage 2x a week (every 3-4 days)

with antibiotics. Gook luck! It's an friendly cell line.

Remember NOT to freeze your HL-60 in DMSO. DMSO will differentiate your cells which some researchers will want to do for their experiments. You have to freeze using GLYCEROL. The only problem with glycerol is that your percentage viability when initiating your cells will go down dramatically.
