GSK3beta inhibitor - searching for good inhibitors of GSK3beta (Mar/14/2007 )

Dear all,

I'm planning to work with a specific inhibitor of GSK3beta for cell culture.
Searching the calbiochem website I found many different inhibitors and as being unexperienced with all these inhibitors, I would like to know if anybody
has some experience with GSK3beta inhibitors and may recommend one or the other (Lithium may be an option aside but not in focus).

Thanks in advance and best regards

You can use LiCl...but Li is not really specific...so you need to use a more specific one like SB compound....I always use the inhibitors by Calbiochem...they are the best in the market....so you can trust their numbers...just make sure you read the details very carefully and pay attention to factors like cell permeability, reversibility, and most importantly the concentration in which it only works aganist GSK3 and not other kinases....otherwise they are all good and I never had any problems with Calbiochem inhibitors....One more suggestion is to go to PUBMED...there you'll find a huge numbers of paper on GSK3B inhibtion....and you can look at the other people resutls to get an idea which one suits your projects...It is always helpful to add one more experiment using GSK3B siRNA pool or dominant negative construct (or adenoviral vectors), if you are thinking on publishing your results in a high impact journal....hope this will help....Good luck

Hi Mitrablue,

thanks for your reply;
I think the siRNA pool would be a good option as well, so I will give it a try.

Thanks,
Cheers

Hi,
I think the SB compunds are what people use (not sure if they are selective for the beta form though). SB415286 should work...
