Simple method for concentrating adeno-associated virus(rAAV) - (Mar/02/2007 )

Hello everyone,
Is there any simple method for concentrating rAAV? Like precipitation or filtration?

Thank you rolleyes.gif

You mean.. by other ways than CsCl purification?

unfortunately there is no simple way to concentrate AAV. Actually its one of the cons for this virus.

HPLC can be helpful, it still not an easy way but atleast better than other methods.

Maybe ultrafiltration by centrigugation with a MWCO 30k-100K be helpful?

Somehow this step never worked for us. Maybe for invivo stuff we needed it to be really concentrated, which wasnt possible using this method.

I had heard that some virus can be precipitated by PEG8000. Can this also apply to rAAV?

I havent come across a paper which uses PEG8000 for AAV for purification and concentration.

or may be I havent followed AAV as closely.

But one can always try different methods.
