cell synchronization - (Feb/22/2007 )

hi guys

I'm studying the effect of a growth factor on the phosphoraylation of several proteins such as erk 1/2. I usually starve my cells in serum free media for 24 hours before the treatment. however not every time i starve my cells i get complete downregulation of the phophorylated proteins. whenever i get high background in my control untreated cells i cannot see the effect of the growth factor and when i get complete downregulation i can see nice time response effect. the question is what should i do to ensure complete downregulation of the prateins im studying....thanks very much for any help

What test do you do to make sure that the serum starvation did indeed result in cell synchronisation? Because in my experience it doesn't neccessarily...
