MCF7 wound healing (scratch assay) - (Jan/31/2007 )

Hi everyone

did anyone do a wound healing experiment using MCF7 ( scratch assay) ? I'm having some trouble getting consistant results sad.gif

is the problem more methodically f.i. to scratch a clear line, or inconsistency in results of migration?

the inconsistancey in the result. when i used 100 nm of the test peptide i obtained good wound healing. when i increased the dose to 1 UM it did not work very well. however i also did it at higher confluency too with the 1 UM dose (100% confluency). I usually do it in serum free conditions too, i dont know if this will affect the result.
