Tissue spots shred from Tissue Microarray Slide - (Jan/28/2007 )

Dear all,

I found some of the tissue spots began shredding out of my tissue microarray slide when I did the washing. I wonder if i did something wrong. Because I put slide in a dish with wash buffer and shake very gently in a orbital shaker.

Any comment? Thank you!!!

Do you retrieve the tissue by heating beforehand? I always use a squirt bottle and run a slow stream of PBS over the tissue, but some tissues are just more susceptible to falling off. Sometimes it depends on how you cut and put them on the slide and what kind of slides you use too. I use Fisher plus slides.

Thx very much for your kind reply.
I bought the tissue microarray slides from Pantomics, Inc. I also followed the protocol they provided.
The first step is Deparaffinization: Bake slides in an oven at 60oC for 30 mins to 1 hour. And then soak the slides in 1. Xylene 2 X 5 minutes
2. 100% ethanol, 2 X 5 minutes
3. 95% ethanol, 1 X 3 minutes
4. 70% ethanol, 1 X 3 minutes
5. Distilled water, 1 X 3 minutes
I used a dish to immerse the slides in the above steps with very gently shaking and some tissues falled off.

I would contact them and see what they say about it. If you followed their protocol, then they should refund your money or give you replacement slides.
