we use 25mM tris-cl ( pH 8.0) and 0.25M sucrose and no triton for subcellular fractionation.

Hello, scolix.
Would you please tell me the detailed method for subcellular fractionation? For membrane and cytosl etc.
I have checked some protocols , but do not know what to choose..

there are various protocols around dependending on the specificity of fractionation; should only one specific phase/organell be isolated or various or as many as possible? think also of corresping assays to measure enrichment and recovery of your subcellular fractions...
